Description

The techniques employed to manipulate mouse embryos require a range of skills from the basic to the advanced. Many of these techniques are used in the manual Manipulating the Mouse Embryo: A Laboratory Manual, Second Edition. This video shows step-by-step demonstrations, with running commentary, of the following techniques:

  1. Dissection of oviducts. Recovery of fertilized eggs. Removal of cumulus cells.
  2. Recovery of embryos. Removal of the zona pellucide. Construction ofaggregation chimeras.
  3. Recovery of blastocysts.
  4. DNA injection into pronuclei to produce transgenic embryos.
  5. Nuclear transfer.
  6. Blastocyst injection of inner cell mass and embryonic stem cells.
  7. Vasectomizing male mice.
  8. Oviduct transfer of manipulated embryos.
  9. Uterine transfer of manipulated embryos.
  10. Dissection of 6-1/2 and 7-1/2 day embryos.
  11. Dissection of midgestation 12-1/2 day embryos and fetal membranes.

The instructors are Roger Pedersen (University of California, San Francisco) and Janet Rossant (University of Toronto), both experts with many years' experience in the development and teaching of these techniques.

Videotaped demonstration has already proved in other professions to be a valuable and effective teaching technique, especially for imparting the subtle tricks that spell the difference between success and frustration.
